Ostreolysin A6 (OlyA6) is a membrane-binding protein derived from the oyster mushroom (Pleurotus ostreatus). It specifically binds to sphingomyelin/cholesterol (SM/Chol) domains or ceramide phosphoethanolamine (CPE) (a major sphingolipid in insect membranes) with nanomolar affinity (KD = 1.3 nM). When combined with its partner protein pleurotolysin B (PlyB), OlyA6 forms 13-subunit transmembrane pores that target and disrupt insect midgut membranes (e.g., in the western corn rootworm), demonstrating potent insecticidal activity.

The E69A mutant of OlyA6 expands its functionality by enabling binding to free sphingolipids without cholesterol, making it a valuable tool for studying lipid rafts. Additionally, OlyA6 interacts with β-tubulin to regulate cytoskeletal functions. Its versatility positions OlyA6 as a promising candidate for bioinsecticide development and membrane biology research.

Further dilutions should be made in appropriate buffered solutions.
