Calcium Independent Sortase A

Sortases are a class of transpeptidases that utilize the thiol group of an active-site cysteine to modify proteins by recognizing and cleaving the C-terminal sorting signal LPXTG (where X is any amino acid) between threonine (T) and glycine (G) residues. Sortase 2A.9 is an engineered variant derived from the wild-type Sortase of Staphylococcus aureus, exhibiting significantly higher activity than the wild-type enzyme. It is used for site-specific labeling of antibodies or proteins containing the LAXTG recognition sequence (where X is any amino acid). This enzyme enables facile attachment of diverse tags, including peptides, DNA, carbohydrates, or fluorophores containing a polyglycine sequence (Gly)ₙ (where n ≥ 3 glycine residues). Sortase 2A.9 is protected under U.S. Patent No. 10,202,593.

Application:

Sortase 2A.9 recognizes antibodies or genetically engineered proteins containing the LAXTG motif (X = any amino acid). The enzyme cleaves this sequence between threonine and glycine residues, after which the terminal glycine is replaced by any polyglycine (G)ₙ tag to conjugate labels such as HRP, biotin, fluorophores, and others. Sortase 2A.9 is compatible with diverse labeling applications and reaction conditions.

Reaction Buffer:300 mM Tris-HCl, 150 mM NaCl, 5 mM CaCl₂, pH 7.5

复溶/Reconstitution:

Before use this product, please read the direction below carefully. This vial must be briefly centrifuged prior to opening to bring the contents to the bottom. Reconstitute in a sterile distilled water or aqueous buffer containing 0.1% BSA to a concentration of 0.1-1.0 mg/ml. Stock solutions should be apportioned into working aliquots and stored at ≤ -20℃. Further dilutions should be made in appropriate buffered solutions.
