mOrange2

mOrange2 is a modified fluorescent protein derived from mOrange through the introduction of specific mutations (Q64H/F99Y/E160K/G196D). It has a molecular weight of 26.8 kDa, an excitation wavelength of 549 nm, an emission wavelength of 565 nm, a maximum excitation efficiency of 0.6, a brightness of 34.8, a pKa value of 6.5, and a half-life of 2.7 seconds.

mOrange2 exhibits higher photostability compared to the original mOrange protein, with an approximately 30% improvement in photostability. Moreover, its photobleaching curve shows that it is more stable than EGFP. In addition, mOrange2 is a pH-sensitive fluorescent protein suitable for measuring intracellular pH values. Its emission spectrum is sensitive to pH changes within the range of pH 6.0 to 8.0.

mOrange2 is widely used in biological research, including cell labeling, protein localization, cell tracking, and bioimaging. It is commonly used in mammalian cell expression plasmids, combined with the CMV promoter for efficient expression of the target protein. mOrange2 is also employed as a biosensor for detecting physiological processes such as NHE3 and DRA activities.

Further dilutions should be made in appropriate buffered solutions.
