 Recombinant Human Interleukin-37

Human Interleukin-1 family member 7 (IL-1F7), also named FIL-1 zeta, IL37, IL1H4, IL-1HL and IL1RP1, is an anti-inflammatory member of the IL1 cytokine family. Alternative splicing generates multiple isoforms (IL37a through e) with deletions in the N-terminal region of the molecule. IL-37b is up-regulated by inflammatory stimuli in peripheral blood mononuclear cells. Experimental over-expression of IL37b in vivo limits the inflammatory response and protects mice from colitis and LPS-induced shock. Both unprocessed and mature IL37b can form homodimers in solution. Although IL37b will bind to IL18 R alpha with low affinity, this has no effect on IL18 receptor activity. Alternatively, IL37b will also bind to IL18BP, generating a complex that interacts with IL18 R beta. This has the effect of attenuating IL18 activity via the IL18 receptor. IL37b can also function intracellularly. Following LPSinduced cleavage of its propeptide, IL37b associates with Smad3 and translocates to the nucleus. This results in a reduction of proinflammatory cytokine secretion. The longest IL-1F7 transcript encodes IL-37b, which shares approximately 21%, 24%, and 30% aa sequence identity with mature IL-1α, IL1β, and IL-1ra, respectively. Mouse IL37 has not been reported, but human IL37b is active on mouse cell. Recombinant IL-37 is a single non-glycosylated polypeptide chain containing 166 amino acids.

Before use this product, please read the direction below carefully. This vial must be briefly centrifuged prior to opening to bring the contents to the bottom. Reconstitute in a sterile distilled water or aqueous buffer containing 0.1% BSA to a concentration of 0.1-1.0 mg/ml. Stock solutions should be apportioned into working aliquots and stored at ≤ -20℃. Further dilutions should be made in appropriate buffered solutions.
